| Description: | The biochemical activity of compounds was determined by incubation with specific enzymes and substrates in the presence 1.4 uM ATP/[gamma-32P] ATP. After incubation, the reaction was stopped and the phosphorylated substrate was separated from nonincorporated radioactive ATP using SPA beads (Amersham Pharmacia), counted using a scintillation counter to measure substrate-incorporated phosphate. Values for pIC50 were obtained using nonlinear regression analysis. |
