| Competitive Displacement Binding Assay |
Description: | The dissociation constant of the hCA and inhibitor complex was determined by competitive displacement of a fixed concentration of dansylamide by increasing concentrations of the inhibitor using Perkin-Elmer lambda 50-B spectrofluorometer. The excitation and emission wavelengths were maintained at 330 and 448 nm. Since the binding of the inhibitor competitively displaced dansylamide from the enzyme site, the fluorescence intensity at 448 nm decreased. The dissociation constant of the enzyme-inhibitor complex was determined by monitoring the decrease in the fluorescence of the enzyme-dansylamide complex as a function of the increasing concentration of the inhibitor. |
