Assay Name:  Enzyme Inhibition Assay
Description:  HIV-1 protease inhibitor activities were determined by the fluorescence resonance energy transfer (FRET) method. The energy transfer donor (EDANS) and acceptor (DABCYL) were labeled at two ends of peptide substrate to perform FRET. Fluorescence measurements were carried out on a fluorescence spectrophotometer. Excitation and emission wavelengths were set at 340 and 490 nm. Inhibitor binding dissociation constant Ki was obtained by nonlinear regression fitting to the plot of initial velocity as a function of inhibitor concentrations based on Morrison equation. The initial velocities were derived from the linear range of reaction curves.
