Assay Method Information

Assay Name:  Serine Protease Inhibition Assay
Description:  The enzyme reactions were initiated by the addition of substrate, and the color developed from the release of p-nitroanilide from each chromogenic substrate was monitored continuously for 5 min at 405 nm on a microtiter plate reader. The initial velocities measured were used to determine the amount of inhibitor required to diminish 50% of the control velocity; this concentration was defined as the IC50 of the inhibitor. The apparent Ki values were then calculated according to the Cheng-Prusoff equation: Ki = IC50/1 + [S]/Km.
