| Description: | Compounds were tested for their ability to inhibit BACE-1 hydrolysis of the internally quenched fluorescent substrate FS-2. Reactions were initiated by the addition of BACE-1 to reaction buffer containing test compound and substrate, and brief mixing. The increase in fluorescence over time was monitored on a Gemini XS fluorometric plate reader using excitation at 328 nm and emission at 405 nm. Initial rates were fit to an equation, and Ki values were determined using Graphpad Prism software. |
