Assay Method Information

Assay Name:  Determination of the Inhibition Type and Constant (Ki)
Description:  The Km and Kappm for the amino acid substrate were first calculated from Lineweaver-Burk plots. The Ki values were calculated from the Kappm vs [I] plot. The Ki values for the inhibitors with respect for the amino acid substrates were determined by measuring the apparent Km for the amino acid in the presence of saturating concentrations of both ATP and tRNA, and of various fixed concentrations of the inhibitor. Curve-fitting of these data using the theoretical functions for vi/v0 for various types of inhibition was made with the software Kaleidagraph (version 4.0) and was used to identify the types of inhibition and the Ki values.
