| | Capsaicin Antagonist Assay |
| Description: | TRPV1 activities were monitored as a function of cellular uptake of radioactive calcium (45Ca2+). The test compounds were preincubated with TRPV1 expressing CHO cells, which were seeded in 96-well plates at a density of 20,000 cells/well two days prior to the assay. Then 45Ca2+ and capsaicin (final concentration, 0.5 uM) were added, and left for additional two min prior to compound washout. Radioactivity in the 96-well plates was measured using a MicroBeta Jet (Perkin-Elmer, Inc.). IC50 values were calculated by generating antagonist inhibition curves using Xlfit version 2.0.6 (ID Business Solutions, Ltd). The reported results were the average of at least two independent experiments with three replicates at each concentration (+/- SEM). |
