| Description: | Assays were performed by incubating aliquots of rat kidney mitochondria suspensions with [3H]PK 11195 and a range of concentrations of the test compounds (1 nM - 10 uM) in binding buffer. Total and non-specific binding was determined in the absence and in the presence of unlabelled PK 11195 (1 uM). The incubation was stopped by sample filtration and the radioactivity was counted using a scintillation counter. Protein concentration of mitochondrial suspensions was estimated in accordance with the method of Bradford. For the active compounds, the IC50 values were determined and Ki values were derived in accordance with the equation of Cheng and Prusoff. |
