Assay Name:  Assay of Enzyme Activity
Description:  The activity was assayed by measuring the rate of change in NADPH fluorescence (at 455 nm with an excitation wavelength of 340 nm) at 298 K. When the fluorescence due to the high concentration of inhibitor interfered with the fluorometric assay, the enzyme activity was determined by measuring the rate of change in NADPH absorbance at 340 nm. The inhibitor constant, Ki, was determined from a Lineweaver-Burk plot using five concentrations of substrate, and is expressed as the mean +/- standard error of at least three determinations.
