Assay Name:  Human Liver GPa Enzymatic Activity Assay
Description:  An enzymatic assay was developed to measure the response of the activated form of glycogen phosphorylase to small molecule compounds. The assay was to monitor the glycogenolytic reaction by coupling the production of glucose-1 -phosphate from glycogen and inorganic phosphate to phosphoglucomutase, glucose-6-phosphate dehydrogenase, NADH oxidase and horseradish peroxidase to produce the fluorescent product resorufin. The assays were performed in duplicate in 96 or 384-well microtiter plates and the change in fluorescence due to product formation was measured on a fluorescence plate reader using a 560 nm excitation filter and 590 emission filter.
