| Description: | Compounds were tested for their ability to inhibit the cleavage of the substrate by the purified enzyme in a fluorescence-based fluorescence resonance energy transfer (FRET) assay. The human TACE protein was pretreated with the inhibitors for 10 min at room temperature. The reaction was initiated by the addition of pro-TNF-alpha peptide to the TACE protein, and the increase in fluorescence was monitored at excitation of 320 nm and emission of 420 nm over a period of 10 min. Under this assay condition, the IC50 value is very close to the Ki value. |
