| Description: | Test compounds were serially diluted in the assay buffer. In each well of a 96-well microtiter plate (Immunofluor B, Dynatech), the inhibitor solution and the diluted enzyme were combined at room temperature. After 10 min, the activity was assayed by adding assay buffer containing substrate, and continuously monitoring the increase in fluorescence (ex@380 nm;em@440 nm) of the hydrolysis product, MCA-Pro-Leu. The enzyme activity was defined as the change in fluorescence value measured in a 6 min interval divided by 6. The IC50 value was determined by plotting % inhibition of activity vs inhibitor concentration. |
