Assay Name:  AChE Inhibition Assay
Description:  AChE inhibitory activity was evaluated spectrophotometrically by the method of Ellman, using AChE from bovine or human erythrocytes and acetylthiocholine iodide as substrate. The 5,5-dithiobis(2-nitrobenzoic) acid (DTNB) solution was used to produce the yellow anion of 5-thio-2-nitrobenzoic acid. Inhibition curves were performed in triplicate by incubating at least 12 concentrations of inhibitor for 15 min. One triplicate sample without inhibitor was always present to yield 100% of AChE activity. The reaction was stopped with 100 ul of 1mM eserine, and the color production was measured at 414 nm.
