| Description: | For determination of IC50 values, HIV-1 protease was added to assay buffer containing inhibitor and the substrate (H-His-Lys-Ala-Arg-Val-Leu- (p-NO2) Phe-Glu-Ala-norleucine-Ser-NH2). The Leu- (p-NO2) Phe bond of the substrate was cleaved by the enzyme, and the substrate and products were separated by reversed-phase HPLC. Absorbance was measured at 220 nm, peak areas were determined, and percentage conversion to product was calculated. |
