| Description: | Enzymatic reactions were initiated by adding kinase to the reaction mixture containing ATP, [gamma-33P] ATP, peptide substrate and test inhibitor compound. The reactions were terminated after 10 min at 23 °C. The reaction mix was then transferred to filter plates for scintillation counting. Each inhibitor compound was added to the reaction to produce an 11-point dose-response curve (0.0001 to 10 uM). The IC50 values were estimated from data fit to the equation: y =Vmax X (1 - (x/(K + x)). |
