Assay Method Information

Assay Name:  Kinase-Glo Assay
Description:  In a typical assay, 10 μL of test compound of different concentrations (dissolved in dimethyl sulfoxide [DMSO] and diluted with assay buffer) and 10 μL (20 ng) of enzyme were added to each well followed by 20 μL of assay buffer containing substrate and ATP to get a concentration of 25 μM substrate and 1 μM ATP per well. The final DMSO concentration in the reaction mixture was <1%. After incubation at 30 °C for 30 min, the enzymatic reaction was quenched with 40 μL of Kinase-Glo reagent. Luminescence was recorded after 10 min using Infinite F200® PRO multimode reader (Tecan Group Ltd., Männedorf, Switzerland). The activity is proportional to the difference of the total and consumed ATP.
