Assay Name:  LoxA Selectivity Assay
Description:  The one-point inhibition percentages were determined for human LOX inhibitors by following the formation of the conjugated diene product at 234 nm (ε = 25,000 M−1 cm−1) at an inhibitor concentration of 25 μM. All reaction mixtures were 2 mL in volume and constantly stirred using a magnetic stir bar at room temperature (22 °C) with 7 nM LoxA. Reactions were conducted in 25 mM HEPES buffer (pH 7.0), 0.01% Triton X-100, and 10 μM AA. The concentration of AA was quantitatively determined by allowing the enzymatic reaction to go to completion. Confirmed hitswere then screened against the human isozymes 5-LOX, 15-LOX-1, 15-LOX-2, and 12-LOX for specificity.
