| Description: | The inhibition assay of these compounds for AChE (electrophorus electricus) and BuChE (equine serum) were determined by the spectroscopic method described by Ellman et al. which was adapted for 96-well microplates. 25 The standard compound used was galantamine. A 75 μL of different concentrations of the sample was dissolved in buffer with a maximum of 10% DMSO, 25 μL of ATCI (15 mM), and 125 μL of DTNB 3 mM were added to the wells followed by 25 μL of 0.3 U/mL AChE. The velocity was measured by the increase in the absorbance at 405 nm during 1 min time interval at 25 C in a BIO-TEK ELX800G microplate reader (using Gen5 v.1.05 software). |
