| Description: | β-Glucuronidase activity was determined by measuring the absorbance at 405 nm of p-nitrophenol formed from the substrate by the spectrophotometric method. The total reaction volume was 250 無. The reaction mixture contained 185 無 of 0.1 M acetate buffer, 5 無 of test compound solution, 10 無 of enzyme solution was incubated at 37 蚓 for 30 min. The plates were read on a multiplate reader (SpectraMax plus 384) at 405 nm after the addition of 50 無 of 0.4 mM p-nitrophenyl-b-D-glucuronide. All assays were run in triplicate. Furthermore to avoid precipitation, the concentration of the compounds was decreased and the reaction volume was high (200 無) so the chance of precipitation was less hence the addition of detergents was not needed. |
