Assay Method Information

Assay Name:  α-Glucosidase Inhibition Assay
Description:  The enzyme assays were done in the initial linear parts of the assay curve with suitable positive and negative controls (data not shown). A total of 100 mL enzyme assay volume contained 70 mL of 50 mM phosphate buffer pH 6.8, 10 mL of 0.5 mM test compound and 10 mL (0.0234 units, Cat. No. G5003, Sigma) of yeast α-glucosidase Type-I enzyme (E.C. 3.2.1.20). After 10 min pre-incubation at 37 °C, the absorbance was measured at 400 nm followed by the addition of 10 mL of 0.5 m p-nitrophenyl-α-d-glucopyranoside (Cat. No. N1377, Sigma). Incubation was continued for further 30 min at given temperature. The absorbance was measured using Synergy-HT, BioTek, USA, 96- well plate reader. All experiments were performed in triplicates.
