Assay Method Information

Assay Name:  CB1 Receptor Binding Assay (In Vitro)
Description:  The target compounds were evaluated using a binding assay towards the CB1 receptor expressed on the membranes of Chinese hamster ovarian (CHO) cells. Rimonabant was used as the positive control and DMSO was used as the blank control. The potent synthetic cannabinoid agonist, [3H]-CP55940, was used as a radioligand (Figure 1). The CHO cells were incubated in 96-well microtiter plates. Various concentrations of the synthesised cannabinoid ligands were added and incubated for 10 min at 37°C. Then the [3H]-CP55940 was added at a final concentration of 1.7 nM and incubated for another 30 min at 37°C. The cells were lysed and scintillation fluid was added. The radioactivity was quantitated by liquid scintillation spectrometry [Menozzi et al., Eur. J. Med. Chem., 43:2627-2638]. There were three parallels in the tests.
