| Description: | PTP1B was diluted before the experiment to 1.2 μg/mL in Tris buffer, pH7.6 (10 mM Tris, 1.0 mM EDTA, 3.0 mM DTT, 0.01% w/v NaN3). The tested compounds were dissolved in DMSO. The enzyme reaction buffer was Tris buffer, pH 7.5 (50 mM Tris, 0.15 M NaCl, 3 mM DTT). A typical 10 μL of tested compounds was added to the enzyme solution (50 μL), and the resulting mixture was preincubated at 37°C for 30 min. The enzyme reaction was initiated by the addition of 8 mM p-NPP (40 μL). The concentration of p-nitrophenol from the reaction mixtures was recorded at 405 nm, 15-min intervals for 120 min. SOV was used as a positive control in this study. |
