| | In Vitro Pancreatic Lipase Assay |
| Description: | Various concentrations of test compound (5, 10, 25, 50 and 100 mM) were dissolved in DMSO (final culture concentration 0.1%) and 4-MU oleate and lipase were diluted in 0.1M McIlvane buffer containing 0.1M citric acid-Na2HPO4 (pH 7.4). The mixedsolution that composed of 100 mL of 0.1mM 4-MU oleate, 40 mL of the above-mentioned buffer and 10 mL of test samples was prepared before adding lipase. The enzymatic reaction was started by adding 0.05mL of 1 U/mL pancreatic lipase and incubation was carried out for 20 min at 37 °C. The amount of 4-methylumbelliferon released by the enzyme was monitored at an emission of 450 nm and excitation wavelength of 320 nm using a fluorescence spectrophotometer. |
