| QC Inhibition Testing |
Description: | QC activity was evaluated fluorometrically using Gln-AMC as substrate, and pyroglutamyl peptidase as the auxiliary enzyme. After conversion of Gln-AMC into pGlu-AMC by QC, the pGlu-AMC was hydrolyzed by pyroglutamyl peptidase. The generated AMC was detected with excitation/emission wavelengths of 380/460 nm. All determinations were carried out using a BMG Novostar reader for microplates. The inhibition constants were evaluated by fitting the data to the equation for competitive inhibition. |
