| Description: | Kd values were obtained by incubating His-PPARgamma-LBD with biotinylated peptide, europium-labeled anti-histidine antibody, and allophycocyanin-labeled streptavidin in black flat-bottom 384-well microplates. Time-resolved fluorescence was measured on an Analyst GT multimode reader (Molecular Devices, Sunnyvale, CA). The results were expressed as the ratio of allophycocyanin fluorescence to europium fluorescence multiplied by 10,000. Kd values were then calculated. EC50 is the concentration of test compounds needed to induce 50% of the maximum luciferase activity in HepG2/C3A cells co-transfected with PPARgamma-LBD and Gal4-luciferase reporter construct. |
