| | TACE Inhibition Assay |
| Description: | Enzyme activity was determined by a kinetic assay measuring the rate of increase in fluorescent intensity generated by the cleavage of an internally quenched peptide substrate. The reaction was started by the addition of the substrate. The fluorescent intensity (excitation at 320 nm, emission at 405 nm) was measured every 45 s for 30 min using a fluorospectrometer (GEMINI XS, Molecular Devices). Values of Ki were calculated by the PRISM program based on one-site competitive inhibition mode. Each Ki value was an average of three determinations, and the standard errors for all Ki determinations were less than 10%. |
