| | Cathepsin B Inhibition Assay |
| Description: | Inhibition of cathepsin B was assayed in reaction buffer (50 mM NaOAc-HCl, 2 mM dithiothreitol, 2 mM EDTA, pH 5.5) containing 20 µM substrate and 1.5 nM cathepsin B. RR-AMC was used as a substrate for cathepsin B. Cathepsin B was activated by incubation in assay buffer at 37 °C for 30 min prior to initiation of the reaction by addition of the substrate and test compound. The reaction mixture was then incubated at room temperature for 30 min with shaking. Fluorescence intensities were measured at 360 nm excitation and 450 nm emission wavelengths. CA074 was used as a positive control for inhibition of cathepsin B. |
