| Description: | Assay Protocol: Pre-Incubation of Z-AAT with Compounds1. 7.5 μL of Z-AAT (20 nM) was incubated with compounds 1-227 in a GCA plate for 1 hour at room temperature. Addition of hNE1. 7.5 ul of HNE solution (3 nM in PBS+0.01% BRIJ35) added into GCA plate2. Incubate plate for 30 minutes to allow Z-AAT/HNE suicide complex formation. Addition of Substrate and Read Plate on PE Envision1. 7.5 μL of substrate (300 μM solution of elastase substrate (ES V) in PBS+0.01% BRIJ35) dispensed per well into GCA plate2. Immediately read on Envision. |
