| In Vitro Assay for Inhibition of Factor Xa |
Description: | The ability of test compounds to inhibit human fXa in vitro was determined in a fluorescence assay using rhodamime 110, bis-(Boc-L-glycylglycyl-L-arginine amide) as the fluorogenic substrate. Test compound and enzyme were preincubated for 15 min prior to addition of the substrate. Incubation was for 3 h, after which a LJL Analyst (now marketed by Molecular Devices Corporation) was used to monitor the fluorescence at 485/535 nm wavelength. Analysis of the derived data using Activity Base, a curve fitting software from IDBS, gave the pIC50 and pKi values from which Ki values were derived. |
