Assay Name:  In Vitro SYK Inhibition Assay
Description:  SYK kinase was purified as a full length protein in a baculovirus system near homogeneity. All kinase assays were performed with the Kinase TK (tyrosine kinase) HTRF (Homogeneous Time Resolved Fluorescence) assay developed by Cisbio international. These assays were carried out at room temperature in 96-wells half-area white plates in a final volume of 25 μl of kinase buffer (10 mM MgCl2; 2 mM MnCl2; 50 mM Sodium-HEPES pH 7.8; BRIJ-35 0.01%, 1 μM substrate) containing ATP at a concentration of at least twice the Km for each enzyme and an appropriate amount of recombinant enzyme to ensure a linear reaction rate. Reactions were initiated upon introduction of the enzyme and terminated with the addition of one reaction volume (25 μl) of HTRF detection buffer. Plates were incubated for one hour at room temperature and the time resolved Fluorescence resonance energy transfer signal was measured in a Pherastar FS microplate reader (BMG Labtech). All data were the average of triplicate results with a standard deviation <10%.
