| | Determination of Inhibitor Ki |
| Description: | The reaction was started by addition of pNPP substrate, and reaction progress was monitored at 405 nm. The initial rate data collected was used for determination of Ki values. For Ki determination, the kinetic values were obtained directly from nonlinear regression of substrate-velocity curves in the presence of various concentrations of inhibitor. Each inhibitor Ki value was determined using at least 6 independent measurements and at least two enzyme concentrations (final concentration of enzyme: 25 nM and 12.5 nM). |
