Assay Name:  In vitro DPP-IV Inhibitory Assay
Description:  DPP-IV inhibitory activity was determined by measuring the p-nitroaniline (pNA) released from the chromogenic substrate hydrolysis (H-Gly-Pro-pNA). The DPP-IV Drug Discovery Kit-BML-AK 499 used for assay was taken from Enzo Life Sciences International, Inc., Plymouth Meeting, PA, USA. The DPP-IV inhibitor P32/98 was selected as standard inhibitor. The assay was carried out in 96-well flat-bottomed microtiter plates. The stock solutions of the test compounds were prepared in DMSO and diluted in buffers (50 mM Tris HCl) to final concentrations ranging from 10 to 1000 µM in the assay. 10 µl of each compounds was added in each reaction well. The final concentration of DMSO in the assay well is 1% whichis found to have no demonstrable effect on DPP-IV enzyme activity.
