Assay Name:  Isothermal titration calorimetry
Description:  Titrations were performed by a 0.4 µl injection followed by 19 successive injections (2 µl) of each compound(adenosine 500 µM, PS653 250 µM) into a 202 µl reaction cell containing PDK150-359 (26.5 µM and 23 µM,respectively). For the titrations of PDK1 with adenosine, the protein and the compound were prepared in 50 mMTris-HCl (pH 7.4), 200 mM NaCl, 1 mM DTT and 1% v/v DMSO. For the titration of PDK1 with PS653 theprotein and the compound were prepared in 50 mM Tris-HCl (pH 7.4), 200 mM NaCl, 1 mM DTT and 5% v/vDMSO. Errors on the thermodynamic parameter values in Table 1 are non linear least square fitting errors of theexperimental binding isotherms using the Levenberg-Markardt iteration method (Freire et al., 2009). The titrationof PDK1 with adenosine was performed at 25 C, and the titration of PDK1 with PS653 was made at 37 C. Rawcalorimetric data were corrected for heats of dilution.
