| | Tryptophan Fluorescence Assay |
| Description: | Tryptophan fluorescence measurements were performed with purified FdTSPO1 protein and ligand essentially as described by Li et al.; 2.5 μM protein in 250 μL of purification buffer con- taining 0.02% DDM with or without 2 mM β-mercaptoethanol was used. Protein was excited at 280 nm and 600 V and emission measured at 290−400 nm with a PTI QuantaMaster spectrofluorimeter (HORIBA Instruments, Edison, NJ). Ligand was added in aliquots until all fluorescence was quenched. |
