Assay Name:  UV-Visible Titration Assay
Description:  All UV-visible spectra were measured on a Varian Cary 50 spectrometer with temperature control from a Peltier cooler. The holo-enzyme was titrated in 0.1M potassium phosphate buffer starting at pH 6.8, 25 °C, and finishing at pH 5.1 or 10.2. Each titration was conducted in a 2-ml cuvette with the protein at ~8 uM heme-containing monomer, where the heme concentration was determined via the pyridine hemochrome assay. Solutions were continuously stirred, and the pH was monitored using a pH meter with a glass electrode (Corning, pH 430). Small volumes of HCl (1 M) or NaOH (1 M) were added to the solution; the pH was measured, and UVvisible spectra were recorded. All spectra were adjusted for dilution.
