| TRPV1 Channel Activity Assay |
Description: | The effect of the substances on Ca2+ influx was determined by using HEK-293 cells stably overexpressing recombinant human TRPV1 cDNA. The cells were loaded with Fluo-4, a selective intracellular fluorescent probe for Ca2+. Experiments were carried out by measuring cell fluorescence before and after the addition of the test compounds at various concentrations. Agonist activity was determined in comparison to the maximum Ca2+ influx due to the application of 4 uM ionomycin. EC50 values were determined as the concentration of test substances required to produce half-maximal increases in [Ca2+]i. All determinations were at least performed in triplicate. Curve fitting (sigmoidal dose-response variable slope) and parameter estimation were performed with GraphPad Prism (GraphPad Software Inc., San Diego, CA). |
