| | AChE and BChE Assay |
| Description: | The AChE inhibition assay was performed according to the Ellman's method. Total volume of 100 無 reaction mixture contained 60 無 50 mM Na2HPO4 buffer, pH 7.7. Ten 無 test compound (0.5 mM well#1) was added, followed by the addition of10 無 (0.005 unit well#1 AChE, 0.5 unit well#1 BChE, Sigma Inc.) enzyme. The contents were mixed, pre-incubated for 10 min at 37 蚓 and pre-read at 405 nm. The reaction was initiated by the addition of 10 無 of 0.5 mM well#1 substrate (acetylthiocholine iodide or butyrylthiocholine chloride, Sigma Inc.), followed bythe addition of 10 無 DTNB from Sigma Inc. (0.5 mM well#1). After 15 min of incubation at 37 蚓, absorbance was measured using 96-well plate reader Synergy HT, Biotek, USA. All experiments were carried out with their respective controls in triplicate. Eserine (0.5 mM well#1) was used as a positive control. |
