| Tyrpsin Amidolytic Assay |
Description: | Buffer and 0.1 mL of enzyme solution was added to 0.2 mL of examined compounddissolved in 0.15 M NaCl (as control 0.15 M NaCl). The buffer and the enzyme solution included: Borane buffer − 0.5 mL (pH 7.5), enzyme: trypsin (0.4units/mL), synthetic substrate: Bzl-L-Arg-pNA HCl (0.2 mL, 8 mM). The mixture was incubated for 3 min at 37°C. After 20 min of incubation, the reaction was stopped by adding 0.1 ml of 50% acetic acid, and the absorbance of the released p-nitroaniline was measured at 405 nm (Spekol 1300, AnalyticJena). |
