Comments: | Principle of the assay Patch clamp is gold standard to measure channel activities. The purpose of this assay is to assess a compound ztz240 (IUPAC name: N-(6-chloropyridin-3-yl)-4-fluorobenzamide; PubChem SID: 87350301 ) for its effects to potentiate KCNQ2 potassium channel. This assay employs manual electrophysiology to investigate the current response of HEK-KCNQ2 elicited by voltage clamp protocols in the presence or absence of test compound. Compounds were tested in 6-plicates at varying concentration. Keywords: KCNQ2, agonist, activator, potentiator, JHICC, Johns Hopkins, Molecular Libraries Probe Production Centers Network, MLPCN. Protocol for testing ztz240 with voltage clamp 1. cDNAs and Mutagenesis The KCNQ1 to KCNQ5 cDNA were gifts from Drs. T. Jentsch (Zentrum fur Molekulare Neurobiologie, Hamburg), D. Mackinnon (State University of New York, Stony Brook), M. Sanguinetti (University of Utah), M. Shapiro (University of Texas Health Science Center, San Antonio) and V | |
