| Description: | The inhibitory activity on the isolated ATM kinase was determined by a commercial supplier (Reaction Biology Corp., PA, US) using an activity based FRET assay. Compounds were therefore first pre-incubated with human ATM before substrate (p53) and ATP were added to initiate the phosphorylation reaction. After a given time, the kinase reaction was stopped and the amount of phosphorylated substrate was quantified using standard FRET detection methods. Based on a 5-point dilution row starting at 10 μM with a 10-fold dilution factor, the half-maximal inhibitory concentration (IC50 value) was calculated for each final compound.
