| Description: | Purified 5-LO was diluted in PBS containing EDTA (1 mM) and freshly added ATP (1 mM) and incubated with the myxochelin test substances for 10 min at 4°C, followed by 30 s at 37°C. Addition of CaCl2 (2 mM) and arachidonic acid (20 µM) initiatedthe 5-LO product formation. After 10 min, the reaction was terminated by adding ice-old MeOH (1 mL), and the production of 5-LO metabolites was analyzed by RP-HPLC. |
