| Description: | The test compounds were dissolved in assay buffer (50 mM Tris, 150 mM NaCl, 10 mM CaCl2, 0.05% Brij35, pH 7.5) at a final concentration of 10 μM and 1 μM. Coagulation factor Xa and test compound were added to the well plate, and incubation was conducted at room temperature for 10 minutes after mixed thoroughly. The substrate (Mca-RPKPVE-Nval-WRK(Dnp)-NH2) was added to initiate the reaction. For reading fluorescence signal value, enzyme kinetic mode was used. The wavelength of excitation light was 320 nm, and the wavelength of emission light was 400 nm. |
