| | Fluorescence Assays for Determination of Binding Affinity. |
| Description: | Binding was detected as a change in the intrinsic tryptophan fluorescence of the PI3K upon the addition of inhibitor. The inhibitor was incubated with the protein for at least 15 min before the fluorescence intensity was measured. Fluorescence data were acquired for 30 s and averaged for each concentration of inhibitor. The background fluorescence of a solution without inhibitor was subtracted from each measurement. Data were then fitted to a binding equation to obtain dissociation constants. |
